Germination Test
Germination % = (number of normal seedlings / total seeds tested) × 100
Conducted on 400 seeds (4 × 100) on moist filter paper or sand at controlled temperature and light conditions specific to each species. Results after the recommended test period distinguish normal seedlings, abnormal seedlings, hard seeds, and dead seeds. Minimum germination standards are set by state and federal seed laws for commercially sold seed.
Seed Vigor Tests
Germination percentage measures maximum potential under ideal conditions. Vigor tests predict field performance under stress. Common vigor tests:
- Accelerated aging (AA): Seeds exposed to high heat and humidity (41°C, 100% RH) for 48–72 h, then germinated — simulates poor storage and field stress
- Cold test: Seeds germinated in cold, wet soil (10°C) to simulate early-season planting stress
- Electrical conductivity: High leakage from seeds in water indicates membrane deterioration and low vigor
Purity Analysis
Determines the percentage of pure seed of the species of interest vs. inert matter, other crop seeds, and weed seeds. Conducted by weighing subsamples and separating components visually. Results guide seeding rate corrections — low purity means more seed must be planted to achieve target plant density.

Only normal seedlings are counted — abnormal seedlings, hard seeds, and dead seeds are classified separately.
Seed vigor measures the ability of seeds to perform well under field stress conditions — not just in ideal lab conditions. Two seed lots may show 90% germination in the lab but very different emergence rates in cold, wet, or dry field conditions. Vigor tests (accelerated aging, cold test, electrical conductivity) predict which lot will perform better under real-world stress. High-vigor seed is especially important for early planting in cold soils, sandy or compacted soils, and in precision agriculture where skips (unfilled row positions) are costly.
Purity analysis determines what percentage of a seed lot is pure seed of the labeled species, vs. inert matter (broken seeds, chaff, soil), other crop seed, and weed seed. Conducted by weighing a working sample, separating components by hand under magnification, and weighing each fraction. Results are reported as: % pure seed, % inert matter, % other crop seed, % weed seed. High weed seed contamination can disqualify a lot from commercial sale and create long-term weed management problems in the field.
Seeding rate (seeds/area) = Target plant density / (germination fraction × purity fraction × expected field emergence fraction). Example: target 30,000 plants/acre, 85% germination, 98% purity, 85% field emergence: seeds needed = 30,000 / (0.85 × 0.98 × 0.85) = 30,000 / 0.708 ≈ 42,373 seeds/acre. Convert to weight using thousand-seed weight (TSW): lb/acre = (seeds/acre × TSW) / 453,592. This ensures adequate seed is purchased and planted to achieve the target stand.
