Lipid unsaturation refers to the number of double bonds (and rings) in a fatty acid or lipid molecule. The degree of unsaturation (or index of hydrogen deficiency, IHD) can be calculated from the molecular formula. For fatty acids, the number of double bonds between carbon atoms determines whether they are saturated (no double bonds, solid at room temperature), monounsaturated (one double bond, liquid), or polyunsaturated (two or more double bonds). Degree of Unsaturation Formula

DoU = (2C + 2 + N − H − X) / 2

C = carbons; H = hydrogens; N = nitrogens; X = halogens. For a pure hydrocarbon (no N, X): DoU = (2C + 2 − H) / 2. Each double bond = 1 DoU; each ring = 1 DoU. Example: palmitic acid C₁₆H₃₂O₂: DoU = (2×16 + 2 − 32) / 2 = 2/2 = 1 (the C=O carboxyl group). Oleic acid C₁₈H₃₄O₂: DoU = (2×18 + 2 − 34)/2 = 4/2 = 2 (1 C=C double bond + 1 carboxyl C=O).

Fatty Acid Classification

  • Saturated: No C=C double bonds; general formula CₙH₂ₙO₂; solid at room temperature (palmitic 16:0, stearic 18:0); animal fats

Iodine Value

The iodine value (IV) measures total unsaturation of a fat or oil: IV = (moles I₂ absorbed × 254 g/mol I₂) / mass of fat. Coconut oil (mostly saturated): IV = 5–10. Olive oil (monounsaturated): IV ≈ 80. Flaxseed oil (highly polyunsaturated): IV ≈ 190. High IV = more unsaturated = more susceptible to oxidative rancidity.

Health Implications

Saturated fats: increase LDL cholesterol. Unsaturated fats: improve lipid profile, reduce inflammation. Trans fats (artificial partially hydrogenated): raise LDL, lower HDL — banned from US food supply (2018).
