Environmental Exposure Pathways
Humans are exposed to environmental contaminants via three main routes: inhalation (airborne particulates, gases, vapors), ingestion (contaminated water, food, or soil), and dermal contact (soil, water, chemicals on skin). Total exposure integrates across all pathways and is expressed as dose = concentration × intake rate × exposure duration / body weight.
Dose-Response Relationships
The dose-response relationship quantifies how the probability or severity of a health effect changes with exposure level. For most carcinogens, a linear no-threshold (LNT) model assumes no safe dose. For non-cancer endpoints, threshold models assume a level below which no adverse effect occurs — the No Observed Adverse Effect Level (NOAEL) is divided by safety factors to set reference doses (RfD) for regulatory purposes.

For carcinogens, regulatory agencies typically assume a linear no-threshold (LNT) response — any dose carries some risk. For non-cancer endpoints, a threshold is assumed below which no adverse effect occurs, quantified by the NOAEL and used to derive health-protective reference doses (RfD).
The Reference Dose (RfD) is the EPA's estimate of a daily oral exposure that is likely to be without appreciable risk of adverse health effects over a lifetime. It is calculated from animal toxicology studies: RfD = NOAEL / (uncertainty factors × modifying factors). Typical uncertainty factors of 10 each account for animal-to-human extrapolation and human variability, giving a total UF of 100–1000. RfD is expressed in mg/kg body weight/day.
PM2.5 refers to fine particulate matter with aerodynamic diameter ≤ 2.5 μm. PFAS (per- and polyfluoroalkyl substances) are a group of thousands of synthetic chemicals used in non-stick coatings, water-repellent fabrics, firefighting foam, and food packaging. They are extremely persistent in the environment and human body ('forever chemicals') due to strong carbon-fluorine bonds. Health concerns include associations with thyroid disease, immune dysfunction, certain cancers (kidney, testicular), and elevated cholesterol.
