Doubling time is a critical parameter in cell biology used to characterize cell lines, optimize culture conditions, and plan experiments that require specific cell densities at defined time points.

How It Works

During exponential growth, cell number increases according to Nf = Ni × 2^(t/Td), where Td is the doubling time. Solving for Td:

Td = t × log(2) / log(Nf / Ni)

The specific growth rate (µ) in natural log units is:

µ = ln(Nf / Ni) / t

The total number of population doublings is:

Doublings = log₂(Nf / Ni)

Frequently Asked Questions

Most commonly used mammalian cell lines have doubling times between 18 and 30 hours. HeLa cells typically double in about 22-24 hours, CHO cells in 14-17 hours, and HEK293 cells in 34-36 hours. Primary cells are usually much slower, often 48 hours or more.

Ensure cells were in exponential growth phase during the measurement period. If cells were in lag phase (just after passaging) or stationary phase (confluent), the doubling time will be inaccurate. Also verify that your cell counts are accurate and that the time measurement is precise.

Growth rate (µ) and doubling time (Td) are inversely related: µ = ln(2) / Td. A higher growth rate means a shorter doubling time. The growth rate is expressed in units of per-time (e.g., per hour) and represents the fractional increase in population per unit time.
